386. EFFECTS OF IN UTERO AND LACTATIONAL EXPOSURE TO 2, 3, 7, TCDD ; ON OVARIAN ARYL HYDROCARBON RECEPTOR LEVELS AND FOLLICULAR ESTRADIOL PRODUCTION. Travis B Salisbury1 and Jennifer L Marcinkiewicz1. 1Kent State University, Dept. of Biological Sciences, Kent, OH. TCDD is a widespread environmental contaminant. In utero and lactational IUL ; exposure of rats to TCDD causes reproductive tract abnormalities, disrupts estrous cycles and ovulation. IUL exposure also reduces follicle numbers and serum estradiol E2 ; levels. Treatment of rat and human granulosa cells with TCDD reduces E2 biosynthesis. The objectives of this study were to evaluate the effects of IUL exposure to TCDD on 1 ; E2 biosynthesis of intact antral follicles 2 ; protein levels of ovarian aryl hydrocarbon receptors AHR ; . On day 15 of pregnancy, rats were treated with TCDD 2.5 g kg n vehicle acetone: corn oil, 19: 1 n 6 gavage. On postnatal day 23, the 15 largest antral follicles were isolated and incubated with FSH 100 ng ml ; , LH medium alone 1ml ; . The medium was collected after 4 hrs of incubation and stored until RIA. Rats on day 23 were also injected subcutanously with PMSG 10 IU ; . hours after PMSG, the 9 largest follicles were collected and incubated for 2 hours. AHR protein levels were evaluated by Western blot analysis of ovaries collected on day 23 and 48 hours post PMSG. Differences between treatment groups were determined by ANOVA. Within isolated follicles day 23 ; , both FSH and LH treatment resulted in a 2 fold increase in E2 levels when compared to basal E2 levels. Follicles collected 48 hours after PMSG also showed significantly greater E2 production. There were no differences in E2 production by follicles from TCDD-exposed animals compared to controls at any time point. On day 23, AHR protein levels were significantly reduced 2.3 fold ; within ovaries of TCDD-exposed animals compared to controls. Interestingly, AHR protein levels in ovaries collected 48 hours post-PMSG were reduced 4 fold when compared to AHR protein levels within ovaries prior to PMSG. There were no significant differences in AHR protein levels between TCDD-exposed animals and controls on day 25. The results of this experiment demonstrate that E2 biosynthesis on a per follicle basis ; is not compromised following IUL exposure to TCDD, suggesting that the reduction in serum E2 levels is most likely a consequence of fewer E2-producing follicles. The mechanism and significance of TCDD-induced reductions in AHR protein levels within the ovary warrant further investigation. 387. PREPUBERTAL EXPOSURE TO 2, 3, 7, TCDD ; IS ASSOCIATED WITH A PREMATURE TRANSITION TO REPRODUCTIVE SENESCENCE IN FEMALE SPRAGUE-DAWLEY RATS. Activation of the aryl hydrocarbon receptor pathway by dioxin-like compounds has been shown to promote mechanisms of reproductive aging acutely but long term studies are lacking. In the current study, female rats n 6 per group ; received a single dose of 2, 3, 7, TCDD, 10 g kg p.o. ; or vehicle on d29 of age. TCDD is a potent activator of the AhR and this dose is within the range of accidental human exposure to the compound. Reproductive cyclicity was assessed until one year of age through examination of daily vaginal cytology for one week per month. Using a rat model system, this study examined the effects of preconception exposure to nicotine on regulation of the estrous cycle and several developmental parameters of the conceptus. Virgin Sprague-Dawley female rats were assigned to four groups, NIC 35 mg nicotine ; , NIC 15 mg ; , PLACEBO 0 mg ; and NORMAL control. Nicotine was delivered as a subcutaneous implanted pellet designed for 21-day timed release. Following nicotine pellet implantation, vaginal smears were conducted daily to determine the stages of estrous cycle. On the 19th day after implantation, each female rat was paired with an untreated male rat. Sperm positive gestational day [GD] 0 ; rats were weighed daily and sacrificed on GD 20. Each fetus was removed from the uterus and several developmental parameters were recorded. These data indicated that preconception nicotine exposure altered the estrous cycle 271 and levorphanol.

Nigel M. Kelly is a postdoctoral researcher in the Grant Institute of Earth Science, University of Edinburgh. He received his PhD in geology from the University of Sydney. His research interests include the tectonothermal evolution of ancient orogenic belts as keys to understanding the behaviour of the continental crust, both now and in the past, and involve field work in places such as Antarctica, Greenland, NW Scotland and central Australia. This work has led to a focus on the behavior of dateable accessory minerals during metamorphism and on how these minerals help us place constraints on the rates of orogenic events and act as tracers of many geological processes. Andreas Mller is a research fellow at the Universitt Potsdam, Germany. He received a diploma in geology from the Christian-AlbrechtsUniversitt in Kiel and in 1996 a doctorate from the same institution for his study of the petrology and geochronology of Palaeoproterozoic eclogites and Pan-African granulites in Tanzania. The radiogenic isotope work was carried out during a year at the Max-Planck-Institut fr Chemie in Mainz. After postdoctoral positions at the University of New South Wales Sydney ; and the Johannes-Gutenberg Universitt in Mainz, he came to Potsdam in 2001. He uses in situ analytical techniques to link textural, geochemical, and isotopic information for the understanding of metamorphic processes, with special focus on high-temperature terranes and liothyronine.
