Although only a few examples of TPP-dependent inborn errors have been reported, it seems probable that other defects in binding of TPP to dependent enzymes will be found. In summary, the data presented here demonstrate a TPPmediated increase in affinity of BCKA dehydrogenase for the a-keto acid that is specific for thiamin-responsive MSUD. Moreover, a defect in the binding of TPP has been implicated by an elevated apparent Km for TPP for the mutant enzyme. The molecular mechanism that leads to the increase in substrate affinity is not clear. It is possible that a TPP-mediated conformational change in the mutant enzyme renders the a-keto acidbinding site more accessible for the substrate. Further studies are required to substantiate this possibility.
